The GT-Digital™ IDH1/IDH2/NPM1 Mutation Detection Assay Kit for the QuantStudio™ Absolute Q™ Digital PCR System is a Research-Use Only (RUO), patent-pending molecular reagent kit containing all primers, probes, and controls for the detection and quantification of common mutations found in the human IDH1, IDH2, and NPM1 genes. This multi-target kit contains one assay solution and two qualitative controls. The assay solution contains a TAMRA probe that targets wild-type sequence at IDH1-R132 and Cy5 probes that target IDH1-R132C, IDH1-R132H, IDH2-R140Q, and IDH2-R172K mutations. The assay solution additionally contains a FAM probe which targets wild-type sequence at NPM1-W288 where common insertions occur and a HEX probe targeting wild-type sequence within the same amplicon. The presence of a short nucleotide insertion in the region of the FAM binding sequence results in a HEX-only positive signal (from the absence of FAM signal), whereas the presence of non-mutated NPM1 results in a FAM+HEX double positive signal. The assay was validated for the detection of the Type A NPM1 insertion (c.863insTCTG) and may also detect, but not discriminate between, Type B (c.863insCATG), D (c.863insCCTG), and other insertions occurring between c.863 and c.866. Rare NPM1 insertion types occurring between c.867 to c.875 may also be detected with the assay, resulting in a FAM-only positive signal (from the loss of HEX signal). However, these insertion types have not been tested. Qualitative controls included in the kit contain a mutated control at 10% variant allele frequency (VAF) and a wild-type control at 0% VAF for all target regions.

The GT-Digital™ KRAS G12-G13 Mutation Detection Assay Kit for the QuantStudio™ Absolute Q™ Digital PCR System is a Research-Use Only (RUO), patent-pending molecular reagent kit containing all primers, probes, and controls for the detection and quantification of common mutations found in the human KRAS gene. This multi-target kit contains one assay solution and two qualitative controls. The assay solution targets the KRAS G12C, G12V, and G12A mutations in FAM; the G12S, G12R, and G13C mutations in HEX; the G12D and G13D mutations in Cy5; and wild-type KRAS G12-G13 in ABY. A 10% mixed mutant (MT) control and wild-type (WT) qualitative control are included in the kit. Contains enough material for 50 reactions.
